Pregabalin and Celecoxib Reduced Need for Morphine After Lumbar Spine Surgery

Together, pregabalin and celecoxib demonstrated pain control benefits following lumbar spine fusion surgery, resulting in a lower morphine intake. The results from this randomized, controlled study will be presented at the 2018 World Congress on Pain, held September 12-16 in Boston, Massachusetts.

The focus of this study was to evaluate the efficacy of preemptive pregabalin in combination with celecoxib on the reduction of morphine needed for pain management 48 hours after lumbar spine fusion surgery. A total of 60 patients were randomly assigned to an intervention arm, receiving pregabalin (150 mg) and celecoxib (200 mg) 1 hour before surgery, or a control arm, receiving a placebo 1 hour before surgery. Patients were monitored for pain, using the Visual Analogue Scale, and quantity of morphine consumption, by the number of patient-controlled analgesia button pushes, for 48 hours postsurgery.

The intervention arm had significantly lower pain scores (P <.05) and fewer patient-controlled analgesia button pushes (P <.05) than the control arm. There was a significant reduction in morphine consumption at both 24 hours and 48 hours postsurgery for the intervention arm (23.17±5.39 mg and41.10±5.40 mg, respectively) compared with the control arm (29.03±4.37 mg and 52.23±9.56 mg, respectively). Reported adverse events of nausea and vomiting were also significantly lower in the intervention arm when compared to the control arm (3.3% vs 16.6%, respectively; P <.05).

The study authors concluded, “Pregapalin combined with celecoxib had a good preemptive analgesia effect after lumbar spine surgery.”
